Spoiler Alert: If you haven’t watched last night’s episode yet, don’t read any further.
Jacob is Jesus. The other guy seen with Jacob in the opening scene is Satan. The island is where they are both allowed to live.
Think about this:
- Jacob and the other guy (let’s just assume I’m correct and call him Satan for now) were dressed similarly but Jacob had a white shirt and Satan had a black shirt.
- Jacob was seen fishing. The fish has long been used as a symbol of Jesus.
- Jacob healed John Locke.
- Jacob doesn’t age, knows how to make an Egyptian rug, and is fluent in many languages, all suggesting he has been around for a long time.
- Jacob lives in a foot statue. There is a reference in Genesis to Satan striking Man's heel.
"he shall bruise your head, and you shall bruise his heel" (3:15). - The answer to the question, "what lies in the shadow of the statue?" was, in Latin, "He who will protect us." Thanks to Lostpedia for finding that one.
- The banter between Jacob and Satan was indicative of good versus evil themes that one might expect in a conversation between Jesus and Satan.
Satan: “Still trying to prove me wrong aren’t you?”
Jacob: “You are wrong.”
Satan: “Am I? They come. They fight. They destroy. They corrupt. It always ends the same.”
Jacob: “It only ends once. Anything that happens before that, it’s just progress.”
Some other things to consider, if my theory is correct:
The smoke monster serves, or is, Satan. Satan can take the form of any dead bodies that arrive on the island. He took the form of Jack’s dad and, later, John Locke. John Locke, Ben Linus, and Sayid were duped by Satan. They helped him carry out his (successful?) plan to kill Jacob.
The smoke monster confronted Mr. Eko twice. The first time it let him go. The second time, it killed him--after he had started constructing a church building.
Jacob’s cabin had been “used by someone else.” That someone else was Satan after taking the form of Jack’s dad.
Everything that has happened on the island is the result of this battle between Jacob and Satan.
My guess is that Jacob/Jesus had a plan of his own that involved Sawyer, Jack, Kate, Juliet, and (especially) Hurley (what’s in the guitar case?), which we’ll learn more about in the final season. I’m looking forward to it.
Update: In the comments, Dave P (an old friend and classics major in college) translated the latin phrase "Ille qui nos omnes servabit" as "The one who will save us all," which makes for an even more striking Jacob = Jesus connection.

You left out the thing Ricardus (Richard) said when asked who is in the statue's base, 'Ille qui nos omnes servabit'. (The one who will save us all).
Translation Napp used from Lostpedia said protect us...but should it be save? Also, it appears that "Satan" has more limitations. He cannot leave the island, but Jacob can leave. And let's not forget the Hebrew significance in the name Jacob...
Servo,servare,servavi,servatum is the verb, most commonly translated as:
to watch over, keep, protect, observe, save
Any of these work in the context of the show, Servabit, is the 3rd person singular future, he/she/it will ____, so ille qui servabit, the one who will watch over/save
I just like save over protect both are valid. Watch over does evoke more of the sheppard image,
hmm Jack Shepard maybe? more allegory?
Ooh! I hadn't thought of that. The Shepard name does take on a new meaning now.
The writers have used names from philosophy and science, now we have some religious symbolism.
That also answers the question of why Ben had to kill Jacob because Satan appears as Locke. Especially when the body of Locke shows up in the cargo box after he had entered the Foot with Ben.
